Mekanisme Reaksi Sintesis O-Karboksimetil Kitosan (O-CMC) dari Kitosan dan Asam Monokloroasetat (MCAA) dengan Katalis NaOH: Studi Komputasi Ab-Initio
RINGKASAN
Kitosan merupakan biopolimer yang banyak digunakan dalam bidang
biomedical, pangan, kosmetik, lingkungan karena memiliki sifat yang
biodegradibel dan biokompatibilitas. Namun kitosan memiliki kelemahan yaitu
kurang larut dalam pH netral. Untuk meningkatkan kelarutannya kitosan
dimodifikasi menjadi Karboksimetil Kitosan (CMC). Modifikasi kitosan untuk
menjadi CMC dilakukan dengan cara alkilasi langsung menggunakan asam
monokloroasetat (MCAA) dan NaOH sebagai katalis untuk penetrasi pelarut.
Penelitian ini telah mempelajari pengaruh NaOH sebagai katalis pada mekanisme
reksi sintesis CMC dengan metode komputasi ab-initio. Tujuan dari penelitian ini
yaitu mempelajari pengaruh NaOH sebagai katalis pada mekanisme reaksi melalui
energi aktivasi dan tahap penentu laju reaksi.
Mekanisme reaksi sintesis CMC dilakukan dengan dua model mekanisme
reaksi pembentukan O-CMC pada gugus fungsi –OH primer dimer kitosan melalui
reaksi SN1 yang mengahasilkan O-CMC-1 dan pada reaksi SN2 yang menghasilkan
O-CMC-2. Struktur molekul yang terlibat dalam mekanisme reaksi (R: reaktan, TSm-n : keadaan transisi, I-m-n : zat antara dan P-m-n : produk) dioptimasi dengan
metode komputasi ab-initio dengan teori dan basis set HF/6-31G(d,p) untuk
mendapatkan struktur geometri teroptimasi dengan energi minimum. Energi
minimum yang didapatkan digunakan untuk membuat grafik PES untuk
menentukan energi aktivasi dan tahap penentu laju reaksi.
Hasil penelitian ini telah diperoleh struktur geometri teroptimasi molekul
reaktan dan produk dengan energi minimum molekul dimer kitosan sebesar
-328,349x104 kJ/mol, MCAA sebesar -180,280x104 kJ/mol, NaOH sebesar
-62,294x104 kJ/mol, O-CMC-1 sebesar -387,820x104 kJ/mol dan O-CMC-2
sebesar -387,842x104 kJ/mol. Kemuadian struktur geometri teroptimasi molekul
kedaan transisi dan intermediet dengan energi minimum molekul TSO-1-1 sebesar
-180,253x104 kJ/mol, TSO-2-1 sebesar -387.903x104 kJ/mol, TSO-3-1 sebesar
-407,774x104 kJ/mol ,TSO-1-2 sebesar -508,613x104 kJ/mol, TSO-2-2 sebesar
-528,449x104 kJ/mol, dan keadaan intermediet adalah molekul IO-1-1 sebesar
-180,240x104 kJ/mol, IO-2-1 sebesar -387,925x104 kJ/mol , IO-1-2 sebesar
-508,618x104 kJ/mol. Katalis NaOH dapat mempengaruhi mekanisme reaksi
sintesis O-CMC, yaitu pembentukan O-CMC melalui reaksi SN2 lebih disukai dan
lebih mudah terbentuk daripada melalui reaksi SN1 dengan energi aktivasinya
masing-masing sebesar 323,321 kJ/mol dan 787,530 kJ/mol dan tahap penentu laju
reaksi pembentukannya masing-masing pada tahap kedua.
SUMMARY
Chitosan is a biopolymer that is widely used in the fields of biomedical, food,
cosmetics, environment because it has biodegradiable properties and
biocompatibility. However, chitosan has a weakness, which is less soluble in
neutral pH. To increase the solubility of chitosan modified into Chitosan
Carboksimetyl (CMC). Modification of chitosan to become CMC was carried out
by direct alkylation using monocloroacetic acid (MCAA) and NaOH as a catalyst
for solvent penetration. The research has studied the effect of NaOH as a catalyst
in the mechanism of CMC synthesis with the ab-initio computational method. The
purpose of this study is to study the effect of NaOH as a catalyst on the reaction
mechanism through activation energy and the rate of determining the reaction rate.
The mechanism of the CMC synthesis reaction is done by two models of the
O-CMC to functional group –OH primary formation reaction in the SN1 reaction
which produces in O-CMC-1 and in the SN2 reaction which produces O-CMC-2.
The molecular structure involved in the reaction mechanism (R: reactant, TSm-n:
transition state, Im-n: intermediate and Pm-n: product) is optimized by the ab-initio
computational method with theory and base set HF / 6-31G (d, p) to get a geometric
structure optimized with minimum energy. The minimum energy obtained is used to
make a PES graph to determine the activation energy and the rate of determining
the reaction rate.
The results of this study have been obtained by optimizing the geometric
structure of reactant molecules and products with a minimum energy of chitosan
dimer molecules of -328,349x104 kJ/mol, MCAA of -180,280x104 kJ/mol, NaOH of
-62,294x104 kJ/mol, O-CMC-1 of -387,820x104 kJ/mol and O-CMC-2 of
-387,842x104 kJ/mol. The geometry structure is optimized for transition and
intermediate molecules with a minimum TSO-1-1 molecular energy of
-180.253x104 kJ/mol, TSO-2-1 of -387.903x104 kJ/mol, TSO-3-1 of
-407,774x104 kJ/mol, TSO-1-2 of -508,613x104 kJ/mol, TSO-2-2 of
-528,449x104 kJ/mol, and intermediate conditions are IO-1-1 molecules of
-180,240x104 kJ/mol, IO-2-1 is -387,925x104 kJ/mol, IO-1-2 is -508,618x104 kJ/mol.
The NaOH catalyst can influence the mechanism of the O-CMC synthesis reaction,
ie the formation of O-CMC through SN2 reaction is preferable and easier to form
than through SN1 with activation energy of 323,321 kJ/mol and 787,530 kJ/mol
respectively and rate determinant formation reactions respectively in the second
stage.
