Respon Histologis Hipokampus Tikus Wistar Stres Fisiologis Terhadap Perlakuan Suplemen Daging Ikan Gabus (Channa striata) Dalam Pakan
ABSTRAK
Stres akibat kekurangan pakan yang diikuti aktivitas berlebihan merupakan faktor
pemicu terjadinya gangguan struktur dan fungsi hipokampus. Jaringan saraf di
hipokampus dapat beregenerasi secara terbatas pasca stres. Kandungan nutrisi pakan
menentukan proses regenerasi pada jaringan saraf di hipokampus. Penelitian ini
dilakukan untuk memperoleh gambaran histologis hipokampus otak setelah
suplementasi daging ikan gabus dalam pakan. Penelitian ini menggunakan 30 ekor
tikus Wistar jantan terdiri atas 5 perlakuan dan 4 kali ulangan. Perlakuan pada
penelitian ini terdiri atas P0: tikus kontrol, perlakuan stres dan diberi pakan tikus
komersial; P1, P2, P3, dan P4 berturut-turut adalah: perlakuan stres dan diberikan
pakan yang disuplementasi daging ikan gabus 5%; 10%; 15%; dan 20%. Penelitian
ini menggunakan Rancangan Acak Lengkap (RAL). Parameter yang diukur pada
penelitian ini adalah luas hipokampus otak, ketebalan girus dentat, ketebalan Cornu
Ammonis 3 (CA3), dan ketebalan CA1. Data yang diperoleh dianalisis menggunakan
Analysis of Variance (ANOVA) pada taraf kepercayaan 95% dan dilanjutkan dengan
uji Duncan dengan taraf signifikansi 5%. Hasil analisis data menunjukkan bahwa
suplementasi daging ikan gabus dalam pakan memberikan pengaruh nyata terhadap
luas hipokampus, ketebalan girus dentat, dan ketebalan CA3, sedangkan terhadap
ketebalan CA1 menunjukkan hasil tidak berbeda nyata. Simpulan penelitian ini
adalah suplementasi daging ikan gabus dalam pakan setelah pengondisian stres dapat
meningkatkan luas hipokampus otak, ketebalan girus dentat, CA3, dan CA1.

ABSTRACT
Stress due to lack of feed that followed excessive activity was a trigger factor caused
damaged of hippocampus structure and function. Nerve tissue in the hippocampus
could regenerate itself after exposure of stress. Nutrient content of feed influenced the
regeneration of nerve tissue in the hippocampus. This research was conducted to
obtain histological of hippocampus brain after supplementation of snakehead fish
meat on the feed. This research used 30 male Wistar mice consisted of 5 treatments
and 4 replications. The treatment in this study consisted of P0: control, namely Wistar
rats were conditioned to stress during the six days that followed the feeding without
supplementation of snakehead fish meat for 14 days; P1, P2, P3, and P4, respectively
Wistar rats were conditioned stress followed by supplemental feeding snakehead fish
meat 5%, 10%, 15% and 20%. The parameters measured in this research is the
hippocampus area of the brain, gyrus dentate thickness, Cornu Ammonis 3 (CA3)
thickness, and CA1 thickness. Data were analyzed using Analysis of Variance
(ANOVA) at the confidence level of 95% and continued with Duncan test with a
significance level of 5%. The results of data analysis showed that supplementation
with cork fish meat on the feed provide comprehensive real effect on the
hippocampus area, the thickness of dentate gyrus and CA3 thickness, while the
thickness of CA1 shows the results were not significantly different. It was concluded
that supplementation with snakehead fish meat on the feed after stress conditioning
has potentially increases brain hippocampus area, thickness of dentate gyrus, CA3
and CA1.
