Struktur Histologi Glomerulus Dan Tubulus Proksimal Pada Ren Mencit (Mus Musculus L.) Jantan Setelah Paparan Ekstrak Etanol Daun Mimba (Azadirachta Indica A. Juss.)

Daun mimba (Azadirachta indica A. Juss.) sering dimanfaatkan sebagai obat tradisional karena mengandung senyawa bioaktif diantaranya azadirachtin, meliacin, gedunin, nimbidin, nimbolides, salanin, nimbin dan valassin. Konsumsi obat tradisional dalam jangka waktu yang lama dapat menimbulkan efek samping salah satunya pada organ ginjal. Penelitian ini bertujuan untuk mengetahui strukur histologi glomerulus dan tubulus kontortus proksimal pada ginjal mencit (Mus musculus L.) jantan setelah paparan ekstrak etanol daun mimba (Azadirachta indica A. Juss.). Penelitian menggunakan Rancangan Acak Lengkap (RAL) yang terdiri atas 2 perlakuan dengan 15 kali ulangan, yaitu K (kelompok kontrol dengan pemberian akuades) dan P (perlakuan ekstrak etanol daun mimba dosis 14 mg/kg BB). Perlakuan diberikan secara oral dengan volume 0,2 mL selama 21 hari. Pemberian pakan dan minum secara ad libitum. Organ ginjal diisolasi, ditimbang dan dibuat preparat histologi dengan tebal sayatan 5 µm menggunakan metode parafin dan pewarnaan Hematoksilin dan Eosin. Variabel yang diamati pada penelitian ini yaitu bobot ginjal, konsumsi air, diameter glomerulus, tebal kapsula Bowman, diameter tubulus proksimal dan diameter lumen. Data dianalisis menggunakan uji t dengan taraf kepercayaan 95%. Hasil analisis menunjukan tidak terdapat pengaruh bermakna (p>0,05) dari pemberian ekstrak etanol daun mimba terhadap bobot ginjal, konsumsi air, diameter glomerulus, tebal kapsula Bowman, diameter tubulus proksimal dan diameter lumen. Kesimpulan dari penelitian ini yaitu pemberian ekstrak etanol daun mimba selama 21 hari masih aman digunakan sebagai obat tradisional.

Neem leaves (Azadirachta indica A. Juss.) are often used as traditional medicine because they contain bioactive compounds such as azadirachtin, meliacin, gedunin, nimbidin, nimbolides, salanin, neem and valassin. Consumption of traditional medicines for long periods can cause side effect on kidney. This study aims to find out the histological structure of glomerular and proximal tubular in the male mice (Mus musculus L.) kidneys after exposure to the ethanolic of neem leaves extract. Completely Randomized Design (CRD) consists of 2 treatment groups with 15 replications was used in this study. The first group is K (only treated with aquadest) and P group (treated by ethanolic neem leaves extrcat at the dosage 14 mg/ kg body weight). The treatment were given orally with a volume of 0.2 mL for 21 days. Feeding and drinking were carried out ad-libitum. At the 22 days, kidney was isolated, weighted and made for histological processed with 5 μm in thickness using paraffin method with Hematoxylin and Eosin staining. The variables observed in this study were kidney weight, water consumption, glomerular diameter, thick of Bowman capsule, diameter of proximal tubule and lumen diameter. Data were analyzed using the t test with a confidence level of 95%. The results of the analysis showed that the treatment has no significant effect (p> 0.05) to kidney weight, water consumption, glomerular diameter, thick of Bowman capsule, diameter of proximal tubules and glomerular lumen diameter. The study showed that the use of ethanolic neem leaves extract for 21 days still safe to be use as a traditional medicine.
