Karakterisasi Secara Morfologi, Pengukuran Kadar Capsaicin Dan Identifikasi Molekular Internal Transcribed Spacer (ITS) Cabai Gendot (Capsicum Sp.) Asal Dieng

Cabai gendot asal Dieng memiliki tingkat kepedasan yang tinggi dan potensial dalam membantu memenuhi kebutuhan cabai di Indonesia. Namun, informasi terkait cabai gendot asl Dieng masih sangat kurang baik secara morfologi, kadar capsaicin dan analisis filogenetika. Tujuan dari penelitian ini yaitu melakukan karakterisasi morfologi, mengukur kadar capsaicin dan analisis identifikasi daerah Internal Transcribed Spacer cabai gendot asal Dieng. Metode karakterisasi morfologi menggunakan panduan IPGRI (1995), pengukuran capsaicin menggunakan metode kromatografi lapis tipis dan analisis filogenetika melalui identifikasi ITS cabai gendot. Hasil karakterisasi morfologi menunjukkan cabai gendot memiliki bulu halus pada batang, mahkota bunga ungu dan biji berwarna hitam. Kadar capsaicin yang diperoleh yaitu 4.862,64 µg/g dengan tingkat kepedasan 77.802,24 SHU. Berdasarkan hasil BLAST dan analisis filogenetika menunjukkan bahwa cabai gendot memiliki tingkat kekerabatan yang paling dekat dengan Capsicum pubescens.

Gendot chili from Dieng has a high level of spiciness and potential to help meet the needs of chili in Indonesia. However, the information regarding the Dieng Gendot Chili is still lacking morphologically, capsaicin content and phylogenetic analysis. The purpose of this study was to carry out morphological characterization, measure capsaicin content and identifying Internal Transcribed Spacer of gendot chili from Dieng. Morphological characterization method using IPGRI guidance (1995), capsaicin measurement using thin layer chromatography and phylogenetic analysis through identification of ITS chili gendot. The results of the morphological characterization showed that the gendot chili has stem pubescens, purple corolla and black seeds. Capsaicin content obtained were 4.862,64 µg/g with capsaicin level were 77,802.24 SHU. Based on BLAST and phylogenetic analysis showed that gendot chili has the closest kinship level to Capsicum pubescens.
