Studi Aktivitas Antioksidan dan Dose Enhancement Factor Nanopartikel CeO2 sebagai Radioprotektor terhadap Radiasi Sinar-X LINAC 6 MV

Nanopartikel CeO2 menarik minat besar untuk dikaji sebagai bahan biomedis karena memiliki sifat fisis dan dan kimia yang unik. Kehadiran ion Ce3+ dan kekosongan oksigen pada permukaan nanopartikel CeO2 menginduksi sifat redoks/ katalitis yang memungkinkan nanopartikel tersebut berinteraksi dengan radikal bebas. Sifat ini membuat nanopartikel CeO2 berpotensi sebagai bahan antioksidan dan radioprotektor. Tujuan dari penelitian ini adalah untuk mengevaluasi aktivitas antioksidan dan kemampuan radioprotektor nanopartikel CeO2. Aktivitas antioksidan dari nanopartikel CeO2 dipelajari melalui pengurangan radikal bebas stabil 1,1 Diphenyl-2-picrylhidrazyl (DPPH) menggunakan spektrometer UV-Vis. Dose Enhancement Factor (DEF) nanopartikel CeO2 ditentukan melalui pengukuran dosis serap radiasi sinar-X LINAC dengan dosis keluaran 200 MU dan energi 6 MV. Potensi nanopartikel CeO2 sebagai radioprotektor dievaluasi berdasarkan nilai DEF dan kemampuan nanopartikel CeO2 melindungi kematian bakteri Echerichia coli akibat radiasi sinar-X 6 MV. Nanopartikel CeO2 memiliki aktivitas antioksidan yang ditunjukan dengan nilai IC50 sebesar 4,4 mg/mL dan nilai DEF kurang dari satu yaitu berkisar antara 0,937-0,984. Nanopartikel CeO2 dengan konsentrasi 0,2 mg/mL dapat meningkatkan viabilitas bakteri hidup sebesar 24,8 %. Hasil ini menunjukkan nanopartikel CeO2 berpotensi sebagai radioprotektor sinar-X LINAC 6 MV.

ABSTRACT
The CeO2 nanoparticles have been great interested as a biomedical materials because its unique physical and chemical properties. The presence of Ce3+ ions and oxygen vacancy on the surface of CeO2 nanoparticles induce the redox catalityc properties which allows the nanoparticles to interact with free radicals. This property made that CeO2 nanoparticles are potentially as an antioxidant and radioprotector material. The purpose of this study was to evaluate the antioxidant activity and the ability of the CeO2 nanoparticles as a radioprotecor. The antioxidant activity of CeO2 nanoparticles was studied using 1,1-Dipenhyl-2-picrylhidrazyl (DPPH) method. Dose Enhancement Factor (DEF) of CeO2 nanoparticles was determined by measuring the absorbed dose of X-ray LINAC 200 MU with energy of 6MV. The radioprotector property of CeO2 nanoparticles was evaluated based on DEF value and the capability of CeO2 nanoparticles to protec E-coli damage induced by X-ray iradiation 6 MV. The results showed that CeO2 nanoparticles posse good antioxidant activity with IC50 of 4.4 mg/mL and the DEF values are in the range 0.937 to 0.984. The CeO2 nanoparticles concentration of 0,2 mg/mL increased E-coli viability up to 24,8 %. These results indicated that CeO2 nanoparticles are potential as radioprotector.
