Aktivitas Antioksidan Ekstrak Flavonoid Bakteri Endofit Daun Pepaya (Carica papaya L.)
RINGKASAN
Penelitian terdahulu berhasil mengisolasi bakteri endofit F1, F2, F3, F4, dan
F5 yang bersimbiosis dengan daun pepaya. Bakteri endofit F5 telah diteliti dan
memiliki aktivitas antibakteri terhadap bakteri E.coli, S.typhi, B.subtilis dan
S.aureus. Pada bakteri endofit F5 belum dilakukan eksplorasi aktivitas antioksidan
dalam bentuk ekstrak flavonoid, sehingga pada penelitian ini dilakukan uji aktivitas
antioksidan, uji flavonoid secara kualitatif dan kuantitatif, dan uji kadar total fenol
ekstrak flavonoid bakteri endofit F5. Penelitian ini bertujuan untuk memperoleh
data kualitatif penapisan fitokimia bakteri endofit F5, memperoleh ekstrak flavonoid
dan kadar total fenolnya, serta memperoleh data aktivitas antioksidan ekstrak
flavonoid bakteri endofit F5 daun Carica Papaya L.
Penelitian dilakukan dalam beberapa tahap, yaitu peremajaan bakteri
endofit F5, pewarnaan gram sebagai konfirmasi fenotifik kultur bakteri F5, produksi
senyawa metabolit sekunder, penapisan fitokimia, ekstraksi senyawa flavonoid
menggunakan pelarut etil asetat yang didahului dengan hidrolisis dengan asam
klorida, penentuan aktivitas antioksidan dengan metode peredaman DPPH, uji total
fenolik, dan uji total flavonoid.
Bakteri endofit F5 berbentuk batang dengan elevasi cembung, berwarna
putih, dan tepian halus yang merupakan bakteri gram positif. Bakteri endofit F5
mengandung metabolit sekunder berupa flavonoid, alkaloid, dan saponin. Didapat
ekstrak flavonoid dari isolat bakteri endofit F5 seberat 1,281 gram dengan total
flavonoid sebesar 295,809 mg ekuivalen kuersetin/g sampel yang memiliki
aktivitas antioksidan dengan nilai IC50 sebesar 86,703 ppm dan total fenol sebesar
303,727 mg ekuivalen asam galat/g sampel
SUMMARY
Previous research has been successfully isolated F1, F2, F3, F4, and F5
endophytic bacteria which are symbiotic in papaya leaf. F5 endophytic bacteria have
been studied and have antibacterial activity against E.coli, S.typhi, B.subtilis and
S.aureus bacteria. In bacteria endophytic F5 antioxidant activity exploration has not
been done. Due to those reasons,this study is conducted the antioxidant activity,
total flavonoids qualitatively and quantitatively test, and total phenol content
flavonoid extract of F5 endophytic bacteria isolates tests. The aim of this study is
to obtain qualitative data phytochemical screening of bacteria endophityc F5,
flavonoid extract and and total phenol,and antioxidant activity content of flavonoid
extract of F5 endophytic bacteria of Carica Papaya L. leaf.
This study was conducted with several stages, rejuvenation of F5 endophytic
bacteria, gram dye as phenotyphic confirmation of culture F5 bacteria, the
production of secondary metabolites, phytochemical screening, extraction of
flavonoid compounds using ethyl acetate solvent preceded by hydrolysis of
flavonoids with hydrochloric acid, the antioxidant activity with DPPH test and
determination of total phenolic and total flavonoides content.
F5 endophityc bacteria have bacillus shape with convex elevation, white
rounded colonies, and entire edges which is positive-gram bacteria. The content of
phytochemical compounds from F5 endophytic bacterial metabolite extracts are
alkaloids, flavonoids, and saponin. Obtained flavonoid extract of 1.281 grams with
a total flavonoids of 295.809 mg ekuivalen quercetin/g samples that have
antioxidan activity with values IC50 of 86.703 ppm and total phenolic of 303.727
mg ekuivalen gallic acid/g samples.
