Studi Perbandingan Aktifitas Inhibisi α-Glukosidase Empat Tumbuhan Obat Indonesia dan Korelasinya Dengan Kandungan Total Flavonoid dan Total Alkaloid
RINGKASAN
Diabetes mellitus merupakan penyakit kelainan kadar glukosa darah yang
menjadi masalah kesehatan cukup serius di dunia. Strategi pengembangan obat
antidiabetes baru dari bahan alam dapat dilakukan melalui 3 pendekatan, yaitu La
Theorie des Signatures, berdasarkan penggunaan secara tradisional dan berdasarkan
kandungan kimia. Daun bawang, daun kersen, daun mengkudu dan daun wortel
memenuhi pendekatan tersebut. Salah satu obat diabetes yaitu inhibitor α-
glukosidase yang berfungsi menjaga kadar gula dalam darah agar tetap dalam
keadaan normal. Golongan senyawa yang bersifat inhibitor α-glukosidase
diantaranya flavonoid dan alkaloid. Namun, belum ada informasi mengenai total
kandungan total flavonoid dan alkaloid pada keempat tanaman tersebut. Tujuan dari
penelitian ini adalah menelaah hubungan aktivitas inhibisi α-glukosidase dengan
kandungan total flavonoid dan total alkaloid.
Penelitian ini meliputi beberapa tahap. Tahap pertama adalah karakterisasi
simplisia, penapisan fitokimia, profil KLT dan maserasi menggunakan pelarut
etanol 96%. Tahap kedua adalah penentuan kandungan total flavonoid dan total
alkaloid serta tahap ketiga adalah uji inhibisi α-glukosidase menggunakan
spektrofotometer UV-Vis.
Hasil dari penelitian ini didapatkan kandungan kimia daun bawang, daun
kersen, daun mengkudu dan daun wortel berdasarkan hasil skrining fitokimia
menunjukkan adanya alkaloid, flavonoid, tanin, kuinon dan steroid pada keempat
tanaman. Kandungan total flavonoid ekstrak daun bawang, daun kersen, daun
mengkudu dan daun wortel secara berturut-turut adalah sebesar 329,1667;
386,1111; 240,4167; 438,4722 mg ek. kuersetin/ g ekstrak. Kandungan total
alkaloid ekstrak daun bawang, daun kersen, daun mengkudu dan daun wortel secara
berturut-turut adalah sebesar 70,97814; 128,9563; 97,59016; 147,0437 mg. ek.
Atropin/ g ekstrak. Nilai IC50 dari uji inhibisi α-glukosidase ekstrak daun bawang,
daun kersen, daun mengkudu dan daun wortel secara berturut-turut adalah 163,277;
23,201; 55,485; 19,784 ppm. Hasil analisis korelasi menunjukkan adanya korelasi
antara kadar total alkaloid dengan aktivitas inhibisi α-glukosidase. Namun tidak ada
korelasi antara kadar total flavonoid dengan aktivitas inhibisi α-glukosidase.xii
SUMMARY
Diabetes mellitus is a disease of blood glucose abnormality that becomes a
serious health problem in the world. Strategy development of new antidiabetic
drugs from natural materials can be done through three approaches, namely La
Theorie des Signatures, based on the use of traditional and based on chemical
content. Leeks, cherry leaf, noni leaf, and carrot leaf meet the approach. One of the
diabetes drug that α-glucosidase inhibitor which functions to maintain blood sugar
levels to remain in normal circumstances. Classes of compounds that inhibit α-
glucosidase enzyme such as flavonoids and alkaloids. However, there is no
information about the total total content of flavonoids and alkaloids in the four
plants. The purpose of this study is to examine the relationship glucosidase
inhibitory activity containing a total of total flavonoids and alkaloids.α-glucosidase
inhibitors keep blood sugar levels in order to remain normal.
This research covers several stages. The first stage is characterization of
simplicia, phytochemical screening, TLC profile and maceration using 96% ethanol
solvent. The second stage is the determination of total flavonoid content and total
alkaloid and third stage is inhibition activity test of α-glucosidase by
spectrophotometry UV-Vis.
The results of this study, based on phytochemical screening of leeks, cherry
leaf, noni leaf and carrot leaf showed the presence of alkaloids, flavonoids, tannins,
quinones and steroids in the four plants. The content of total flavonoids of leeks,
cherry leaf, noni leaf and carrot leaf extract in a row were 329.167; 386.1111;
240.4167; 438.4722 mg eq. Quercetin / g extract. The content of total alkaloid of
leeks, cherry leaf, noni and carrot leaf extract in a row were70.97814; 128.9563;
97.59016; 147.0437 mg eq. Atropine / g. Extract. IC50 values inhibition test of α-
glucosidase of leeks, cherry leaf, noni leaf and carrot leaf extract in a row are were
163.277; 23.201; 55.485; 19.784 ppm. The result of correlation analysis shows
correlation between total alkaloid content and α-glucosidase inhibition activity.
However, there was no correlation between total flavonoid content and α-
glucosidase inhibition activity.
