Efek Pemberian Ekstrak Etanol Daun Mimba (Azadirachta indica A. Juss.) terhadap Struktur Ren Tikus Putih (Rattus norvegicus L.) yang Diberi Pakan Tinggi Lemak

norvegicus L.) yang Diberi Pakan Tinggi Lemak. Di bawah bimbingan Sri

Pakan tinggi lemak adalah pakan yang mengandung lemak dengan konsentrasi
tinggi. Konsumsi pakan tinggi lemak dalam jangka waktu lama dapat menimbulkan
akumulasi lemak salah satunya organ ginjal. Daun mimba sering dimanfaatkan
sebagai obat tradisional karena mengandung senyawa bioaktif diantaranya
azadirachtin, saponin, tanin, terpenoid, alkaloid dan flavonoid yang diduga
memiliki efek dalam menurunkan kolesterol. Penelitian bertujuan untuk menguji
efek pemberian ekstrak etanol daun mimba (Azadirachta indica A. Juss.) terhadap
struktur ren tikus putih (Rattus norvegicus L.) yang diberi pakan tinggi lemak.
Penelitian menggunakan Rancangan Acak Lengkap yang menggunakan 24 ekor
tikus yang dibagi menjadi 6 kelompok, yaitu P0 (kelompok kontrol negatif), P1
(kelompok kontrol positif diberi pakan tinggi lemak dan kuning telur bebek), P2
(kelompok diberi pakan tinggi lemak dan kuning telur bebek + simvastatin), P3, P4,
dan P5 (kelompok diberi pakan tinggi lemak dan kuning telur bebek + ekstrak
etanol daun mimba dosis 75 mg/gBB, 100 mg/gBB, dan 125 mg/gBB). Organ ginjal
diisolasi, ditimbang dan dibuat preparat histologi dengan tebal sayatan 5 µm
menggunakan metode parafin dan pewarnaan Hematoksilin Eosin. Variabel yang
diamati yaitu bobot ginjal, diameter glomerulus, diameter kapsula Bowman dan
tebal ruang Bowman. Data dianalisis dengan one way ANOVA pada taraf
kepercayaan 95%. Hasil penelitian menunjukan tidak terdapat pengaruh bermakna
(p>0,05) dari pemberian ekstrak etanol daun mimba terhadap bobot ginjal, diameter
glomerulus, diameter kapsula Bowman dan tebal ruang Bowman pada ginjal tikus
yang diberi pakan tinggi lemak. Kesimpulan penelitian ini yaitu ekstrak etanol daun
mimba mampu memperbaiki struktur ginjal tikus hiperlipidemia.

(Azadirachta indica A. Juss.) on the Kidney Structure of White Rat (Rattus

A high-fat diet is a food that contains high concentrations of fat. Consumption of a
high-fat diet for a long time can cause fat accumulation, one of which is kidney
organs. Neem leaves are often used as a traditional medicine because they contain
bioactive compounds including azadirachtin, saponins, tannins, terpenoids,
alkaloids and flavonoids which are thought to have effects in decreasing the
concentration of cholesterol. This study aimed to examine the effect of ethanol
extract of neem leaf (Azadirachta indica A. Juss.) on the kidney structure of white
rat (Rattus norvegicus L.) fed with a high-fat diet. The study used a Completely
Randomized Design that used 24 rats which were divided into 6 groups, namely P0
(the negative control group), P1 (the positive control group was fed with a high-fat
diet and duck egg yolk), P2 (the group was fed with a high-fat diet and duck egg
yolk + simvastatin), P3, P4, and P5 (the group was fed with a high-fat diet and duck
egg yolk + ethanol extract of neem leaf at a dose of 75 mg/g body weight, 100 mg/g
body weight, and 125 mg/g body weight). The kidney organ was isolated, weighed,
and made for histological processed with 5 µm in thickness which used the paraffin
method and Hematoxylin Eosin staining. The variables observed were kidney
weight, glomerular diameter, Bowman capsule diameter, and thickness of Bowman
space. Data were analyzed by one way ANOVA at 95% confidence level. The
results showed no significant effect (p> 0.05) from the administration of ethanol
extract of neem leaf on the kidney weight, the glomerular diameter, the Bowman
capsule diameter, and the thickness of Bowman space in the kidney of rat fed a
high-fat diet. This research concluded that the ethanol extract of neem leaf can
repair the structure of the hyperlipidemic rat kidney.
